Sotos syndrome is caused by mutations within or deletions encompassing a gene called NSD1.
Usually we need two copies of NSD1 to grow and develop, but in Sotos syndrome only one of these copies is working properly. This is known as haploinsufficiency of one of the NSD1 copies.
