At present, there are no disease-specific therapies for CLCN3-related neurodevelopmental disorder. Management is supportive, symptom-directed, and requires a multidisciplinary approach.
Neurology
• Anti-seizure medications for individuals with epilepsy, with regular follow-up for treatment adjustment.
• Brain MRI as clinically indicated to monitor structural changes or progression.
Developmental care
• Early intervention programs, including physical, occupational, and speech therapy, tailored to individual needs.
• Special education services to address learning difficulties and maximize developmental potential.
Behavioral care
• Behavioral therapies for mood and attention difficulties.
• Psychiatric evaluation and treatment when indicated (e.g., for anxiety, self-injurious, or behavioral dysregulation).
General medical care
• Routine pediatric or primary care follow-up, including monitoring of growth and nutrition.
• Management of feeding difficulties or other supportive care needs, if present.
Genetic counseling
• Recommended for families to clarify inheritance patterns, recurrence risk, and implications for family planning.
• Testing of at-risk family members may be indicated depending on the identified variant.
Surveillance
• Periodic neurological assessments (development, seizures, tone, coordination).
• Ongoing monitoring for behavioral concerns.
• Regular developmental evaluations to guide therapy and school planning.
