The disease is caused by a deficiency of ENPP1, an enzyme that cleaves ATP to AMP and pyrophosphate at the cell surface level. Pyrophosphate is the main inhibitor of calcification, and thus its deficiency leads to calcium build up. Over 40 mutations in ENPP1 have been reported.
